can i take the timing cover of when the engine is filled with oil?

If it's the timing belt cover then yes and if it's the rocker cover then yes, but wouldn't turn it over though.

It's a chain on a Xflow. You can take it off though.

If Xflow, then you will need to drop the sump a little bit, which will mean new gaskets, so you might as well drain the oil & take the sump off.
