The tyres will be directional so the spots will got to the left or right side of the car. Sorry I don't know which side.

if there directional ie 48s it will say which direction they should be put in.the spots on side are usually to match up with spots on rims of new cars so the tyre and wheel are balanced .why its not on all rims i dont know .the dot is usually on the outside but check direction arrrow of tread

On bike tyres the dot lines up with the valve to give the best balance option so you don't need so many weights. There should be an arrow for rotation shouldn't there?

Red dot usually marks the high point of the tread and there may be a rim mark on the wheel which marks the low point. These are best aligned. Yellow dot is lightest point on the tyre and should be lined up with valve. If both dots present red dot takes precedence (assuming the rim is marked).
